Résumé : La fonction du GABA dans la maladie d’Alzheimer : preuve de dysfonction et potentiel de cible thérapeutique pour le traitement des symptômes comportementaux et psychologiques de la démence
La maladie d’Alzheimer (MA) est caractérisée par des perturbations dans de multiples neurotransmetteurs importants. Même si de nombreuses études ont tenté d’établir si l’acide gamma-aminobutyrique (GABA) est perturbé ou non chez les patients de la MA, les résultats varient. Nous examinons les preuves de perturbations du GABA chez les patients de la MA, et suggérons que les résultats variés reflètent les sous-types de la maladie qui se manifestent possiblement cliniquement par différents symptômes comportementaux. L’acide gamma-aminobutyrique, principal neurotransmetteur inhibiteur, est depuis longtemps la cible des anxiolytiques, des sédatifs hypnotiques et des anticonvulsivants. Nous examinons l’utilisation clinique des agents du GABA pour traiter les personnes présentant des symptômes de la MA. Quoique des médicaments du GABA de nouvelle génération soient maintenant offerts, ils doivent encore être évalués chez les patients de la MA.
